Twisted gastrulation Protein (TSG) is a secreted, cysteine-rich BMP binding protein which has a role in dorsal/ventral patterning in Drosophila and Xenopus by regulating BMP signaling. TSG functions as an agonist for BMP signaling by controlling the inhibitory actions of the BMP antagonist, Chordin/Sog, and the cleavage properties of the metalloprotease, xolloid/tolloid. Twisted Gastrulation Protein (TSG) is structurally related to the BMP antagonists Chordin an Noggin. TSG N-terminal domain binds BMP protein directly and displays BMP antagonist activity. It can inhibit BMP activity by binding directly to BMP proteins, and can act either as a BMP4 agonist or antagonist (depending on the specific biochemical environment) by binding to the BMP4/Chordin complex. Recombinant human TSG (aa 26-223) produced in CHO cells is a single, glycosylated, polypeptide chain of 198 amino acids, having a molecular mass ranging from 35-43 kDa on SDS-PAGE due to glycosylation. Recombinant TSG has been purified by proprietary chromatographic techniques.
