IL-4 is a pleiotropic cytokine produced by activated T cells and a ligand for the Interleukin-4 receptor. The Interleukin-4 receptor also binds to IL-13, which may contribute to many overlapping functions of this cytokine and IL-13. STAT6, a signal transducer and activator of transcription, has been shown to play a central role in mediating the immune regulatory signal of this cytokine. This gene, IL-3, IL-5, IL-13, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL-13. IL-4, IL-13 and IL-5 are found to be regulated coordinately by several long-range regulatory elements in an over 120 kilobase range on the chromosome. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. IL-4R produced in Sf9 cells is a single, glycosylated polypeptide chain (26-232 a.a.) fused to an 8 aa His-tag at the C-terminus. It contains a total of 215 amino acids and has a molecular mass of 24.7 kDa. IL-4R shows multiple bands between 28-40 kDa on SDS-PAGE under reducing conditions and has been purified using proprietary chromatographic techniques.
